    RNA silencing, a deeply ingrained regulatory mechanism in plant systems, is instrumental in fine-tuning gene expression and in countering the detrimental consequences of both abiotic and biotic stressors. Gene silencing is fundamentally driven by the interplay of Dicer-like (DCL) proteins, Argonaute (AGO) proteins, and RNA-dependent RNA polymerase (RDR), and substantial research has been conducted on their corresponding gene families within plants. Still, the adzuki bean’s genes and their responses to stresses are not adequately characterized.

    Identification of 11 AGO, 7 DCL, and 6 RDR proteins was followed by phylogenetic analyses, which showed their clustering into six, four, and four clades, respectively. To further validate the expression patterns of these genes, quantitative RT-PCR was used on resistant or susceptible adzuki bean cultivars exposed to drought, bean common mosaic virus, and Podosphaera xanthii infections. Their varied responses to abiotic and biotic stresses underscore the specialized regulatory mechanisms of these proteins.

    In adzuki beans, 24 DCL, AGO, and RDR family genes were identified, and an exploration of their sequence characteristics, encoded protein structures, evolutionary links to orthologs in other legumes, and expression profiles under drought and biotic stress was undertaken.     Accumulated data highlights the arcuate nucleus (ARC) of the hypothalamus as a probable mechanism underlying the association of chronic stress, depression, and diabetes. Chinese herbal formula Xiaoyaosan (XYS) significantly enhances depressive-like behavior and glucose tolerance, although the underlying mechanism is currently unknown. Leptin’s impact on the arcuate nucleus (ARC), orchestrated by the leptin receptor (LepR), contributes to both energy expenditure regulation and the modulation of depressive states. We therefore formulated the hypothesis that XYS could potentially influence depressive-like behaviors and glucose intolerance through the leptin signaling cascade, encompassing the LepR-STAT3/PI3K pathway, in the ARC.

    A rat model manifesting depressive-like behavior and susceptibility to glucose intolerance was established using chronic unpredictable mild stress (CUMS) over six weeks. For six weeks, XYS (2224g/kg) was orally administered, and the positive control group received fluoxetine (20mg/kg). Using the open field test (OFT), sucrose preference test (SPT), and forced swim test (FST), depressive-like behaviors were quantified. Fasting blood glucose (FBG) and oral glucose tolerance tests (OGTT) were carried out to determine the influence of XYS on blood glucose. Employing an enzyme-linked immunosorbent assay and an automated biochemical analyzer, peripheral leptin and blood lipids were, respectively, quantified. Using quantitative real-time PCR and western blotting, researchers detected the influence of XYS on the LepR-STAT3/PI3K pathway.

    The administration of XYS resulted in a reduction of CUMS-induced depressive-like behaviors and an increase in blood glucose. XYS showed an increase in the food consumption rate, but its influence on body weight was minimal. The very essence of care quality is defined by the patient’s experience; inadequate labor support and the perception of loss of control during labor are recurring elements in cases of childbirth-related post-traumatic stress disorder (CB-PTSD).     A multicenter cohort study was undertaken in a French perinatal network, using auto-questionnaires as a tool. Women’s perceptions of disrespect during childbirth, as per the Mother’s Caregivers’ Behavior – Satisfaction Questionnaire (BMC-SQ), were recorded at both 3 days and 2 months following birth to determine a principal outcome. Using the Post-Traumatic Checklist Scale (PCLS) and the Edinburgh Postnatal Depression Scale (EPDS), respectively, CB-PTSD and PPD were assessed two months following childbirth.

During the third day following childbirth, 81.3% (10 from a sample of 123) experienced disrespect during childbirth.
