National PID registries offer useful insights on the epidemiology, diagnosis, and natural history of these disorders. In 1999, the Italian network for primary immunodeficiencies (IPINet) was established. We report on data collected from the IPINet registry after 20 years of activity. A total of 3352 pediatric and adult patients affected with PIDs are registered in the database. In Italy, a regional distribution trend of PID diagnosis was observed. Based on the updated IUIS classification of 2019, PID distribution in Italy showed that predominantly antibody deficiencies account for the majority of cases (63%), followed by combined immunodeficiencies with associated or syndromic features (22.5%). The overall age at diagnosis was younger for male patients. The minimal prevalence of PIDs in Italy resulted in 5.1 per 100.000 habitants. Mortality was similar to other European registries (4.2%). Immunoglobulin replacement treatment was prescribed to less than one third of the patient cohort. Collectively, this is the first comprehensive description of the PID epidemiology in Italy.Embryonic development is of great importance because it determines congenital anomalies and influences their severity. However, little is known about the actual probabilities of success or failure and about the nature of early embryonic defects. Here, we propose that the analysis of embryonic mortality as a function of post-fertilization time provides a simple way to identify major defects. By reviewing the literature, we show that even small initial defects, e.g., spatial cellular asymmetries or irregularities in the timing of development, carry with them lethal effects in subsequent stages of embryogenesis. Although initially motivated by human study, in this contribution, we review the few embryonic mortality data available for farm animals and highlight zebrafish as a particularly suited organism for such a kind of study because embryogenesis can be followed from its very beginning and observed easily thanks to eggshell transparency. In line with the few other farm animals for which data are available, we provide empirical evidence that embryonic mortality in zebrafish has a prominent peak shortly after fertilization. Indeed, we show how subsequent mortality rates decay according to a power law, supporting the role of the early embryonic mortality peak as a screening process rapidly removing defective embryos.The default mode network (DMN) reflects spontaneous activity in the resting human brain. Previous studies examined the difference in static functional connectivity (sFC) of the DMN between eyes-closed (EC) and eyes-open (EO) using the resting-state functional magnetic resonance imaging (rs-fMRI) data. However, it remains unclear about the difference in dynamic FC (dFC) of the DMN between EC and EO. To this end, we acquired rs-fMRI data from 19 subjects in two different statues (EC and EO) and selected a seed region-of-interest (ROI) at the posterior cingulate cortex (PCC) to generate the sFC map. We identified the DMN consisting of ten clusters that were significantly correlated with the PCC. By using a sliding-window approach, we analyzed the dFC of the DMN. Then, the Newman’s modularity algorithm was applied to identify dFC states based on nodal total connectivity strength in each sliding-window. In addition, graph-theory based network analysis was applied to detect dynamic topological properties of the DMN. We identified three group-level dFC states (State1, 2 and 3) that reflects the strength of dFC within the DMN between EC and EO in different time. The following results were reached (1) no significant difference in sFC between EC and EO, (2) dFC was lower in State2 but higher in State3 in EC than in EO, (3) lower clustering coefficient, local efficiency, and global efficiency, but higher characteristic path length in State2 in EC than in EO, and (4) lower nodal strength in the precuneus (PCUN), PCC, angular gyrus (ANG), middle temporal gyrus (MTG) and medial prefrontal cortex (MPFC) in State3 in EC. These results suggested different resting statuses, EC and EO, may induce different time-varying neural activity in the DMN.Taxi and for-hire vehicle (FHV) drivers are a predominantly immigrant population facing a range of occupational stressors, including lack of workplace benefits and increasing financial strain from tumultuous industry changes and now COVID-19’s devastating impact. Bilingual research staff surveyed 422 New York City taxi/FHV drivers using a stratified sampling approach in driver-frequented locations to examine drivers’ health and financial planning behaviors for the first time. Drivers lacked health insurance at double the NYC rate (20% vs. 10%). Life insurance and retirement savings rates were lower than U.S. averages (20% vs. 60%, 25% vs. 58%, respectively). Vehicle ownership was a significant predictor of health insurance, life insurance, and retirement savings. Compared to South Asian drivers, Sub-Saharan African drivers were significantly less likely to have health insurance and North African, and Middle Eastern drivers were significantly less likely to have retirement savings. Although most drivers indicated the importance of insurance and benefits, less then 50% understood how to use them.
